Hygromycin B, Streptomyces hygroscopicus, Concentrated Solution

Description: Hygromycin B, an aminoglycosidic antibiotic produced by Streptomyces hygroscopicus, is used for the selection and maintenance of prokaryotic and eukaryotic cells transfected with the hygromycin resistance gene, hph. Hygromycin B kills bacteria, fungi and higher eukaryotic cells by inhibiting protein synthesis. The resistance gene codes for a kinase that inactivates Hygromycin B through phosphorylation. Cloning of the resistance gene and fusion with eukaryotic promoters has resulted in the development of vectors that permit selection for resistance to Hygromycin B in both prokaryotic and eukaryotic cells.

General Comments: The working concentration for mammalian cell selection is normally between 50 ug/ml and 1mg/ml, Plant cells: 20-200 ug/ml, Bacteria: 20-200 ug/ml and Fungi: 200 ug-1mg/ml. Your appropriate concentration should be tested experimentally.
To determine the Hygromycin B concentration in the 1 MU (Million Units) vial use the following formula. This example is from a previous lot of Hygromycin B. Use the current data provided with your product.
For the 1,000,000 Unit size:
1,000,000 Units at 464,600 U/ml
Volume = 2.15 ml
2.15 ml x 1150 mg/ml = 2,472.5 mg total weight (Hygromycin B + H20)
2,472.5 mg total weight x 37% (% of solids) = 914.825 mg Hygromycin B
914.825 mg Hygromycin B / 2.15 ml = 425.5 mg Hygromycin B / ml
